You need to close your project and re-import it with the desired build configurations. The checkboxes are meant for a future feature to be able to add/removed build configurations, but this is not supported in Express.

Timm-ah's answer applies to the situation where you want to use a config that you did not foresee when you created the project.

If you want to change the build target to one that you enabled when you created the project, you have 2 options (both of which are discussed in the documentation, btw :-):
    - you can choose the build target using the drop down menu in the project Properties -> C/C++ Build
    - using the "i" (small letter i) in the Build configuration window instead of the checkboxes, this will open a drop down to choose configuration as well

The Symbian OS import (Import -> Symbian Bld.inf or Symbian MMP file) will allow you to select build configs based on the SDKs that you have installed, enable the SDKs and target that you need then. Do you have a valid SDK installed, check Windows -> Preferences -> Carbide.c++ -> SDK preferences.
